چکیده
Objectives. To investigate the expression of IL-34 in labial salivary glands (LSGs) of patients with primary SS (p-SS) and its role in inducing a pro-inflammatory monocyte phenotype. Methods. LSG biopsies were obtained from 20 patients with p-SS and 10 patients with non-Sjögren’s sicca syndrome (n-SS). The expression of IL-34, IL-1b, TNF-a, IL-17 and IL-23 was assessed by real-time PCR. IL-34 expression was also investigated in LSGs by immunohistochemistry. The frequencies of subpopulations of CD14 monocytes were evaluated by flow cytometry among isolated mononuclear cells from peripheral blood and salivary glands from both patients and controls. The role of recombinant IL-34 on isolated peripheral blood mononuclear cells was also evaluated. Results. IL-34 m-RNA was overexpressed in the inflamed salivary glands of p-SS and associated with increased expression of TNF-a, IL-1b, IL-17 and IL-23p19. The increased expression of IL-34 was confirmed by immunohistochemistry in paraffin-embedded salivary glands from p-SS patients. IL-34 expression was accompanied by the expansion of pro-inflammatory CD14CD16 monocytes in the salivary glands. In vitro stimulation of peripheral blood mononuclear cells with IL-34 induced the expansion of both CD14CD16 cells and CD14CD16 cells in p-SS and non-SS subjects. Conclusion. IL-34 seems to be involved in the pathogenesis of salivary gland inflammation in p-SS.

Objective(s): To review the findings of the patients with Sjögren’s syndrome (SS) having technetium-99m-pertechnetate (99mTc-pertechnetate) and gallium-67 citrate (Ga-67) salivary gland scintigraphy in the past eight years. Methods: The patients with SS, who were referred to our department for salivary gland scintigraphy during January 2008-December 2015 were studied using both 99mTc-pertechneta...
متن کاملEffect of Shiga Toxin And Its Subunits On Cytokine Induction in Different Cell Lines
Shiga toxins (Stxs) are bacterial virulence factors produced by Shigella dysenteriae serotype 1 and Escherichia coli strains. Stxs are critical factors for the development of diseases such as severe bloody diarrhea and hemolytic uremic syndrome. Additionally, Stxs trigger the secretion of pro- inflammatory cytokines and chemokines, particularly in monocytes or macrophages. The inflammatory cyto...
متن کاملIL-17 is Aberrantly Overexpressed Among Under-treatment Systemic Lupus Erythematosus Patients
Background & Objective: Systemic lupus erythematosus (SLE) is an autoimmune disease with chronic inflammatory immune response. Current therapies mostly rely on glucocorticoids which are accompanied by side-effects and mostly fail to achieve a favorable remission. Th17 subpopulation of T cells is increased in exacerbated SLE as IL-17 cytokine is overexpressed. ...
متن کاملHydrocortisone Reduces Toll-Like Receptor 4 Expression on Peripheral CD14+ Monocytes in Patients Undergoing Percutanoues Coronary Intervention
Bacground: Evidence from several lines of investigations suggests that Toll-like receptor 4 (TLR4) is involved in atherosclerosis as a bridge between innate and acquired immunity. Percutaneous coronary intervention (PCI) can trigger inflammation through activation of human TLR4 (hTLR4) on monocytes. Hydrocortisone as an anti-inflammatory and immuno-suppressant agent has multiple mechanisms of a...
